چکیده
The present study is to investigate the effect of the active fraction of Flos Daturae (AF-FD) (dried flower of Datura metel L.) on IFNinduced apoptosis in a human keratinocyte cell line (HaCAT), which is a widely accepted mimic model of psoriasis. Accordingly, we used MTT and LDH to assay cell viability, FCM and Hoechst 33342 staining to assay cell apoptosis, and Western blotting to assay the expression of keratin 17 (K17). Administration of IFN(5-125 U/mL) decreased MTT reduction and increased LDH leakage in a dose-dependent manner. Twenty-five U/mL IFNelevated apoptosis significantly. The experimental results showed that AF-FD at the concentrations of 25 and 50 μg/mL alleviated the IFN-induced inhibition of MTT reduction by 12.58% and 17.51%, and decreased LDH leakage by 14.47% and 15.83%, respectively. Through FCM and Hoechst 33342 staining, it was found that AF-FD can lower IFNinduced apoptosis at the concentrations of 12.5-50 μg/mL. The administration of 25 μg/mL AF-FD attenuated the IFNinduced over-expression of K17 from 206.32% to 178.83%. The effect of AF-FD on K17 expression may play a key role in protecting HaCAT cells from apoptosis.

Inulae Flos and Its Compounds Inhibit TNF-α- and IFN-γ-Induced Chemokine Production in HaCaT Human Keratinocytes
The present study is to investigate which kinds of solvent extracts of Inulae Flos inhibit the chemokine productions in HaCaT cell and whether the inhibitory capacity of Inulae Flos is related with constitutional compounds. The 70% methanol extract showed comparatively higher inhibition of thymus and activation-regulated chemokine (TARC/CCL17) in HaCaT cells, therefore this extract was further ...
متن کاملPorphyra-334, a mycosporine-like amino acid, attenuates UV-induced apoptosis in HaCaT cells.
The main aim of the current research was to study the effect of porphyra-334, one of mycosporine-like amino acids (MAAs), well known as UV-absorbing compounds, on UVinduced apoptosis in human immortalized keratinocyte (HaCaT) cells. Due to their UV-screening capacity and ability to prevent UV-induced DNA damage, MAAs have recently attracted considerable attention in both industry and research i...

BACKGROUND Calcipotriol ointment has been demonstrated to be a very safe and effective topical drug for psoriasis. This study aims to investigate the effect of calcipotriol on IFN-γ-induced keratin 17 (K17) expression in a human keratinocyte cell line (HaCaT), which is a widely accepted as a mimic in vitro model for psoriasis. MATERIAL AND METHODS We used Western blot, immunofluorescence staini...

متن کاملPropolis Inhibits UVA-Induced Apoptosis of Human Keratinocyte HaCaT Cells by Scavenging ROS
Propolis is a resinous material collected by honeybees from several plant sources. This research aimed at showing its protective effect against UVA-induced apoptosis of human keratinocyte HaCaT cells. Using Hoechst staining, it was demonstrated that propolis (5 and 10 μg/mL) significantly inhibited the apoptosis of HaCaT cells induced by UVA-irradiation. Propolis also showed the protective effe...
